I've got it, and seen it several times. I agree with the general audience and critic consensus that its one of the best animated films ever made. As for it being the "best anime ever", that for me would be a tie between Spirited Away and Akira.

The cel-animation used was amazing, and the themes incorporated in Akira including power, friendship, corruption etc were portrayed very well. I also loved the cyber-punk and gritty/hostile atmosphere.

The last half-hour of the film is particularly awesome.

I've got a theory on the ending line.
At the end, Kaneda (or was it someone else, can't remember) comments on how it has all "yet to begin" (the rebuilt of the world, or the search for more answers regarding the mystery of Akira or whatever). Then Tetsuo is being heard saying "I am Tetsuo."
That line is most likely the one he used to introduce himself to Kaneda in school, thus in a way the line that set everything in motion (if he had never met Kaneda the events in the film would've probably never happened).
So that line is a symbolic "set-up" for these new events.

Perfect Blue is so trippy and haunting, I love it. Grave of the Fireflies is possibly one of the most gruelling and saddest films ever...I got pretty depressed after watching it.

Ouch, definitely a hard call to make out of those two.
Howl's Moving Castle is pretty good, too.

I don't think Akira is the best anime ever, as I don't watch anime a great deal, so I don't know much that I can compare it with. However, I will say that it is one of the best anime movies that I have seen.
